What is Semaglutide

  • Type: Synthetic GLP-1 (glucagon-like peptide-1) receptor agonist.

  • Class: Incretin mimetic / peptide therapeutic.

  • Molecular formula: C₁₇₉H₂₇₁N₄₅O₅₅

  • Molecular weight: ~4113 Da

  • Structure: 31-amino-acid peptide with modifications to prolong half-life:

    • Substituted at position 8 with Aib (α-aminoisobutyric acid) to resist DPP-4 degradation.

    • Conjugated to C-18 fatty diacid chain via a spacer, which binds albumin and prolongs circulation.

 Mechanism of Action

Semaglutide mimics the natural GLP-1 hormone, which is secreted by intestinal L-cells in response to food. It acts through:

  1. GLP-1 receptor activation (pancreas, brain, GI tract)

  2. Enhances glucose-dependent insulin secretion from β-cells.

  3. Suppresses glucagon secretion (when glucose is elevated).

  4. Slows gastric emptying, reducing postprandial glucose spikes.

  5. Reduces appetite via CNS effects, contributing to weight loss.

Essentially, it improves glycemic control while also helping with weight management.
