Melanotan II Acetate – Overview
Melanotan II (MT-II) is a synthetic cyclic peptide derived from the naturally occurring hormone α-melanocyte-stimulating hormone (α-MSH). It was originally developed for photoprotection (reducing UV-induced skin damage) but later gained attention for its broader effects on pigmentation, sexual function, appetite, and energy balance.
The acetate refers to the salt form used to improve stability and handling, not the biological activity itself.
Mechanism of Action
Melanotan II works by activating melanocortin receptors (MCRs), particularly:
MC1R → stimulates melanin production in melanocytes
MC3R & MC4R → involved in appetite regulation, energy metabolism, and sexual behavior
Unlike Melanotan I, MT-II is non-selective, which explains its wider range of physiological effects.
Primary Biological Effects
Skin Pigmentation
Increases eumelanin production, leading to darker skin pigmentation
Enhances tanning independently of UV exposure
May offer some degree of UV protection due to higher melanin levels
Sexual Function
Strongly associated with increased libido and sexual arousal
Effects are mediated mainly through MC4R activation
This property led to the development of bremelanotide (PT-141), a derivative used clinically
Appetite & Weight Regulation
Can cause appetite suppression
May contribute to fat loss via central melanocortin pathways
